hERG Inhibition Assay Service

Screening for the potential risk of QT interval prolongation is necessary to protect patient populations from potentially fatal complications which can be induced by novel pharmaceutical compounds. One of the most influential proteins involved in the control of the cardiac action potential is the product of the human ether-a-go-go gene (hERG). In order to quickly identify compounds with the potential for serious cardiac complications it is recommended that all novel pharmaceuticals be screened for activity against this key gene product before first administration to human volunteers. You can test IC50 by five concentration at late stage, also one or two concentrations at primary screening. So, contact our representative to discuss your needs and ask for report template

hERG Recombinant Cell Line:

CHO-K1 and HEK293 cells are transfected to stably overexpress functional hERG (Ether-a go-go Related Gene). Cells are functional for use in patch-clamp assays.

Our cell lines have been functionally validated using electrophysiological techniques. Predicted sensitivity to known pharmacological agents (Dofetilide, Cisapride, and Terfenadine) has been demonstrated. All pharmacological values are comparable with data previously reported in peer-reviewed journals.

Action Potential Duration Assays (APD)

Action potential recordings reflect the integrated activities of the multiple ion channels, pumps, and exchanger mechanisms that contribute to the depolarization phase and repolarization process at the cellular level. Therefore, assessing changes in action potential duration can be a comprehensive approach for identifying effects of a test substance on repolarization. When a positive effect on one of the action potential parameters is observed in an in vitro assay, ionic current studies should be conducted to elucidate the cellular mechanism for the effect.
